Done today: Red light retest. 4 transformations: Cph8 + pPL-PCB Cph8 PCB -ve control Protocol: 2μL DNA RU1012 30 min ice 1 min 42°C 1 hour at 37°C + LB Each transformation plated onto 2 plates, one of which is wrapped in foil. Placed in 37°C room at genetics which light on Plates: Amp Kan Chlor Arabinose (2mM) IPTG (1mM) X-gal Alex
