Lab Update 10th August
This Report is a summary for full details see the lab notebook
Aims For Friday
- Finish Stage three Ligations (With the exception of the aiiA part)
- Finish Stage two ligations (by wedensday)
- Test part T9002 (This part is the AHL/GFP assay and the Predator AHL sensor)
Aims For today
- Maxiprep and electroporate for ligations John SY
- Retry PCR Cre Lox part John C
- PCR Immunotag Deepti and johhny
Current Ligation Status
- Tick = Done and tested
- ? = Ligated and untested
- X = Done and Failed we will retry
Issues
Riboswitch
- Unable to tell if worked from PCR
- Will be sequenced (takes 2-3 days) waiting on result
PCR
- The PCR results do not correlate with the miniprep results
- PCR for immunotag and lox part is not working
minipreps
- These are not conclusive for many parts, we are looking into using a restriction digest
