Making Glycerol Stocks

Supplies Needed:

  • 60% autoclaved glycerol
  • Day/overnight culture
  • 1.5mL microfuge tube/cryovial

Steps:

  1. Label the tube with: strain, date, BioBrick part number/plasmid or other identifying information, antibiotic (if necessary)
  2. Aseptically add 0.25mL 60% glycerol to the tube, then add 0.75mL of culture (final glycerol concentration 15%).
  3. Vortex briefly.
  4. Put in -80°C freezer.
