Overnight Cultures Prepared 4 overnight cultures in order to make competent cells of BW27783 and DH5α and to miniprep the chlor and amp construction plasmid Innoculated 5mL of LB broth with 09/05/13 BW27783 cells Innoculatd 5mL of LB broth with 09/05/13 DH5α cells Innoculated 5mL of LB-amp broth from a plate of DB3.1 cells containing the Amp construction plasmid Innoculated 5mL of LB-chlor broth from a plate of DB3.1 cells containing the chlor construction plasmid All cultures were grown overnight in a shaking incubator at 32°C
