Ligation Trial 2 Transformation experiment done on 11/08/11 failed. We decide to increase the concentration of DNA in Ligation reaction. We increse concentration to 25 times. It means 1 part of vector and 25 parts of insert. After completing experiment, we realized that for first ligation experiment we have frozen ligation mix.So thats why ligation trial 1 failed.
