    This work was carried out to adapt "building block" approaches for the generation of phosphopeptides through resine-supported solid-phase peptide synthesis to the spot-technology, which relies on cellulose as support. It compares different coupling strategies for the incorporation of protected phoshoaminoacids into determined positions of synthetic peptides. We find that the use of EEDQ (caution, neurotoxic!) as coupling activator is the most efficient strategy in our comparison.

Science in the city

This is a short series of high content essays about the every day life
of scientists in berlin. If you really have the time, or your boss is
not around at the moment, then read ahead and enjoy.

ATTENTION: technical problems (work) are keeping me away from my mission but i will return tu fun as soon as possible.

DAY 1 - ANALYTE CAPTURING


        was just returning from an international symposium and, naturally, the first thing i did once in berlin was to go to the lab and get some very interesting things done. It was not necesary to do it right away, but my sense of responsability was taking over me. At the moment, i did not know that this desition was about to change my whole carreer.

        But i only made it to the stairs because a bunch of squared-headed * guys intercepted me on the way. They were all very disappointed about the attitude of scientists, saying: "these people are always thinking about cheap prices and bonus".

        As it happened: they all belonged to the provider-rebel-army and wanted me to go with them on a special mision. I thought they were quite funny, so i complied. Since then, they have been treating me very nicely. And i have total freedom of speech, as you can see. They took this nice fotograph in the case anybody would be missing me. To call their attention, they said.

          _*_squared-headed_*_ adopted from the spanish "cabeza cuadrada". You would apply this to describe someone who is dyed-in-the-wool or in someway mulish or stubborn. Please do not confuse with its in US narrowly extended use to pejoratively refere to germans. Although there may be cabezas cuadradas in between, I just love the most of you!


DE >> war gerade wieder von einem internationalen Symposium zurück und, natürlich, das erste, was ich einmal in Berlin gemacht habe, war ins Labor zu gehen um einige sehr interessante Dinge zu erledigen. Es war nicht notwendig, es so unmittelbar zu tun, aber mein Gefühl der Verantwortung war stärker als ich. In diesem Moment wusste ich nicht, dass diese Entscheidung dabei war, meine ganze Karriere zu lenken.

Aber ich habe es nur bis zur Treppe geschafft, da eine Reihe von Viereck-köpfigen Jungs mich auf den Weg fing. Sie waren alle sehr enttäuscht über die Haltung der Wissenschaftler. Sie sagten: "Diese Menschen denken immer nur über günstigen Preisen und Bonus".

So wie es geschah: sie alle gehörten zu den Provider-Rebellen-Armee und wollten, dass ich mit ihnen auf eine besondere Mission gehe. Ich dachte, sie wären ganz lustig und war also einverstanden. Seit dann behandeln sie mich sehr gut. Und ich habe die totale Rede-Freiheit, wie man sehen kann. Sie haben dieses schöne Fotograph aufgenommen, falls irgendjemand mich vermissen sollte. Um ihre Aufmerksamkeit zu locken, sagten sie. <<



DAY 2 - INTO THE SKY OF BERLIN


        The results of this week are astonishing. Firstly, I have found that not every squared-headed is really what I supposed to. I can report of 20 little guys, which like to stick covalently together, and a bottle of acetonitrile. I am sure that they are not squared-like at all. My new friends also complained about the way they have been treated: forced to remain on a fixed table for matters of order, to be bathed in acids and toxic solvents. Acetonitrile said that this was true. Indeed, she herself was forced on repeated times to deprive them from water. Every time acetonitrile came, they had to rapidly elute from there. What followed after this was a plethora of tortures.

        It is difficult to tell you about all I’ve seen, I won’t. It would not be fair to upset you in such a way with the sorrow. Instead I can tell you that we have organized ourselves and we are not going to take it anymore. We have broken out on Friday with the fabulous help of the nice Katja. She happily agreed to help us but resisted to come along. Katja didn’t know how her cell cultures would survive the whole struggle which was waiting in the city for all which dare to runaway. From the beginning we were quite aware about the risks, but were determined to be free. She gave us farewell and a smile as she threw us into the sky of Berlin.

DE >> Die Ergebnisse dieser Woche sind erstaunlich. Erstens, ich habe festgestellt, dass nicht jeder Viereck-Köpfige wirklich das ist, was ich bis jetzt angenommen habe. Ich kann von 20 kleinen Jungs berichten, die gerne kovalent zusammenhängen, und von eine Acetonitril Flasche. Ich bin‘s mir sicher, dass sie überhaupt nicht Viereck-artig sind. Meine neuen Freunde beschweren sich auch über die Art, wie sie behandelt worden sind: gezwungen starr auf irgendwelche Tabellen zu posieren, gebadet in Säuren und giftige Lösungsmittel, usw. Acetonitril, sagte, dies sei wahr. Tatsächlich, sie selbst war wiederholt gezwungen, ihnen das Wasser zu entziehen. Jedes Mal, dass Acetonitril kam, mussten sie schnell von dannen eluieren. Was dann folgte, war eine Vielzahl von Folterungen.

Es ist schwer darüber, was ich gesehen habe, zu erzählen. Und ich werde es nicht tun. Es wäre nicht fair, Euch in einer solchen Art und Weise mit der Trauer zu verstimmen. Stattdessen kann ich Euch sagen, dass wir uns selbst organisiert haben, und wir sind nicht mehr zu nehmen. Wir sind am Freitag ausgebrochen, mit der fantastischen Unterstützung der schönen Katja. Sie war bereit uns zu helfen, aber wiederstand unsere bitte mit uns zu kommen. Katja wusste nicht, wie ihre Zellkulturen den gesamten Kampf überleben würden, die in der Stadt auf alle wartete, die es wagen, zu fliehen. Von Anfang an waren wir sehr wohl über die Risiken bewusst, aber auch stärker bestimmt, frei zu sein. Sie gab uns Abschied und ein Lächeln, als sie uns in den Himmel von Berlin warf. <<



DAY 3 - MATTERS OF AFFINITY


        Falling back to Berlin was a very regarding experience. Along the way, the amino acids, acetonitrile and I have had a lot of time. And we are getting to know each other. You would probably say that they are of very simple nature. Just because of the under-microscope dimensions? Well, I will try to explain you how wrong this rapid assumption is.

        I firstly became buddies with acetonitrile. She looks and feels like a princess, and because of the actual narrow pass in her production leading to a devoted appreciation in the market, she is really sexy. It is extremely difficult to explain how this idea got into my head. But at the first moment our eyes crossed, I had the feeling that there was something inside the bottle. So I asked her diverse questions of the small-talk league, just to scrabble around the essence of her being. And I shouldn’t have done this. Not in my condition of a busy scientist earning his degrees. Since then, from the very moment I thought she opened her lid, all theories, hypothesis and axioms are gone from my head. So much place might just be about enough to hold the clear beauty inside acetonitrile.

        I was determined to break all social barriers that were impeding me to fall in love with her. But disadvantageously, during this short period, which passed as we were still on free-fall, something peculiar and notoriously odd about the behaviour of the amino acids drew our attention from ourselves and focused it on them. Our minds were excited by the unexpected facts revealed to us. But this explanation about the real complex nature of the amino acids would blow the frame of this report... for the moment.
