Description

Part 1

  • Prepare 3 sets of hydrogels with phosphate buffers

A)0.51 g of PVOH (MW:89,000-98,000) with 4 ml of phosphate buffer was placed on to a heat block at 100C.

B)0.25 g of PVOH (MW:89,000-98,000) with 3 ml of phosphate buffer was placed on to a heat block at 100C.

C)0.14 g of PVOH (MW:89,000-98,000) with 3 ml of phosphate buffer was placed on to a heat block at 100C.

  • Add 2μ of R6G were added for each ml of phophate buffer. Add 6μ R6G in to B and C. Add 8μ to A.
  • Homoginize each of the hydrogels and place them into freezer. (3 freeze thaw cycle)


Part 2

  • 75ml of fixant solution was placed on a hot plate (stir well). When temperature reaches 60C,500μl of gluteraldehyde was added to the solution. Place 5 small hydrogels (MW:89,000-98,000) (prepared on http://openwetware.org/wiki/User:Samiye_Yaman/Notebook/Lab_581/2013/02/01 ) were placed in to the solution. Every 15 minutes, one of the hydrogels was removed from the solution and placed in to a beaker with distilled water.(keep each hydrogels separate).
