Objective

Make standardized curves for determining peptide concentration using fluorescence analysis.

Procedure

We followed the protocol in Dr. Hartings' notebook. The following are detailed steps we took:

  • Protease Sample Prep.
  1. Used eppendorf tube no. 13 that weighed 1.02669g, and contained 0.00124g of trypsin.
  2. Added 1mL of phosphate buffer.
  3. Final concentration: 53.22µM
  • Sample Prep.
  1. In a 1mL eppendorf tube add:
    1. 0.9812mL of lysozyme solution
    2. 0.0188mL of trypsin solution
  • Bank Prep. (no lysozyme)
  1. In a 1mL eppendorf tube add:
    1. 0.9812mL of buffer solution
    2. 0.0188mL of trypsin solution
