Competent cells

ABSTRACT

  • End of competent cells preparation.

  • We pulled out of the fridge the bottles we left yesterday and centrifuged them again for 10 min at 6000rpm and 4°C. We dilute the pellet in 2 mL of CaCl2 solution 100mM and added also 1 mL of 40% glycerol.
  • Then we transferred 200μL of the content of the bottles to Eppendorf tubes, so we obtain many tubes with cells. We storage the tubes in a jar and inside a fridge that is at -80°C.
  • The cells can be used tomorrow in the afternoon.
