Reaction of OPD and H2O2 in MeOH with Mb/Citrate Catalyst

  • Objectives:
  • 1. Analyze the reaction of O-phenylenediamine and hydrogen peroxide catalyzed by Mb in KCl/Citrate



  • 1. Added 18 mg of O-phenylenediamine to 18 mL of MeOH and vortexed
  • 2. Added 15 mg of Mb lyopholized in KCl/Citrate to 1.5 mL MeOH and vortexed
  • 3. Added 1.134 mL of 30% hydrogen peroxide to 8.866 mL MeOH
  • 4. Centrifuged the Mb solution at 13,200 rpm for 10 minutes
  • 5. The spectrum was taken at 5C, and the H2O2 solution was added at 1% spectrum completion
  • 6. Spectra were then taken at 15C, 25C, 35C, and 45C
  • 7. The data was then analyzed and the spectra were converted to absorbance and corrected
  • Graphs:



  • Observations:
  • 1. Condensation during the 5 Centigrade spectrum - non-useful data
  • 2. The baseline was much higher during the 15C spectrum - will revisit during data analysis
  • 3. Condensation on the 25C spectrum - take note
  • 4. The spectra had different behaviors and peaks between 300 and 800 nm - need to determine what is being synthesized at these peaks
