Making HAuCl4 and Lysozyme Stocks

  • Final [H+] wanted: 0.25 mM
  • Actual amount of HAuCl4: 0.04550 g
    • (slightly more water added to the volumetric flask)
  • Final [Lysozyme] wanted: 250 μM
  • Actual amount of Lysozyme:
  • 50 mL of 0.25 mM HCl was made as a control

Next, solutions were made with gold:protein ratios of 10:1 to 60:1. (See the table). The test tubes were placed in at 80°C for three hours, starting at 13:00.

Kinetics

0.5 mL of HAuCl4 stock and 0.166 mL of Lysozyme stock were added to three test tubes for a 30:1 gold to protein ratio. 4.33 mL of 80°C water was added. Kinetics runs were done every 30 minutes with 2.5 mL of sample in plastic cuvette from 800-360 nm. Started at 13:35.
