Tris buffer preparation

Objective

Two buffer solutions of 10mM Tris+50mM NaCl and 10mM Tris+1M NaCl at Ph of 8.5 were prepared

Procedure

  1. 1.12g Tris and 2.9g NaCl were added to 1L of distilled water to prepare 10mM Tris+50mM NaCl solution and 1.12g Tris and 58.4g NaCl were added to 1L of distilled water to prepare 10mM Tris+1M NaCl solution
  2. Ph meter was calibrated with the standard pH solutions (pH= 4, 7, 10) and afterwards pH of these two Tris buffer solutions were adjusted to 8.5 by adding concentrated 1M HCl
