*Addition of FeSO<sub>4</sub>*7H<sub>2</sub>O to the added Au caused the immediate formation of AuNPs, as indicated by an immediate color change from yellowish tint to a deep/rich purple. This is due to the fact the Fe immediately reduced the free gold in solution.

Objectives

  1. Prepare 10 solutions of varying ratios of BSA, gold, and magnetite to determine the concentrations necessary to prepared BSA fibers with gold and magnetite nanoparticles incorporated into the protein fibers.
  2. Prepare 3 standard BSA and gold nanoparticle protein fibers for later comparison.


BSA-AuNP Preparation


General Protocol:

  1. Prepare a stock solution of HAuCl4 in a volumetric flask using deionized H2O.
  2. Prepare a stock solution of BSA in a volumetric flask using deionized H2O.
  3. Add the appropriate volume of BSA and gold in a 10mL test tube to obtain the desired ratio of BSA:Au. The total volume of the solution should not exceed 5mL.
  4. Add deionized H2O to the test tubes until the solution is 5mL.
  5. Cap the test tubes with either aluminum foil or an aluminum cap.
  6. Heat in oven at 80°C for 3hrs.
  7. Store the solution in a blue falcon tube.

Note:Use caution when removing the test tube from oven. The test tubes will be hot.

Preparation of BSA-AuNP-Magnetite solutions

  • These solutions were prepared following the same method as the BSA-AuNP solutions. These preparation techniques will be compared.
  • The following concentrations of FeSO4*7H2O, KOH, and KNO3 in each prepared solution was adapted from the following #reference[[1]]


General Protocol:

  1. Prepare a 0.3M stock solution of FeSO4*7H2O in a 10mL volumetric flask using deionized H2O.
  2. Prepare a 1M stock solution of KOH in a 10mL volumetric flask using deionized H2O.
  3. Prepare a 0.8M stock solution of KNO3 in a 50mL volumetric flask using deionized H2O.
  4. Add the appropriate volume of BSA and gold in a 10mL test tube to obtain the desired ratio of BSA:Au. The total volume of the solution should not exceed 5mL.
  5. Add the appropriate volume of FeSO4*7H2O to each test tube so that the solution has a concentration of 0.03M FeSO4*7H2O.
  6. Add the appropriate volume of KOH to each test tube so that the solution has a concentration of 0.1M KOH.
  7. Add the appropriate volume of KNO3 to each test tube so that the solution has a concentration of 0.4M KNO3.
  8. Add deionized H2O to the test tubes until the solution is 5mL.
  9. Cap the test tubes with either aluminum foil or an aluminum cap.
  10. Heat in oven at 80°C for 3hrs.
  11. Store the solution in a blue falcon tube.
