Objective

Making new catalase and Hb AuNP samples, as described here for AA and UVVis analysis.

Description

I made the following solutions:

  1. HAuCl4
    1. 24.6mg
    2. 25.0 mL water
  2. Hemoglobin
    1. 15.2mg
    2. 10.0 mL water
  3. Catalase
    1. 40.8mg
    2. 10.0 mL water

I made HbAuNP samples similar to this procedure. Also, there are two ways to think about the ratio of Au:Hb. Hb is a tetramer. So, the molar mass is either 16125 (for the monomer) or 64500 (for the tetramer). The ratios listed below are for the tetramer/monomer. The ratios as reported in the referenced notebook page are those for the monomer.

  1. Au:Hb Ratio = 30/7.5
    1. 500uL gold
    2. 1766uL Hb
    3. 2734uL water
  2. Au:Hb Ratio = 50/12.5
    1. 500uL gold
    2. 1059uL Hb
    3. 757uL water
  3. Au:Hb Ratio = 70/17.5
    1. 500uL gold
    2. 757uL Hb
    3. 3743uL water
  4. Au:Catalase Ratio = 90/22.5
    1. 500uL gold
    2. 589uL Hb
    3. 3911uL water
  5. Au:Catalase Ratio = 110/27.5
    1. 500uL gold
    2. 482uL Hb
    3. 4018uL water
  6. Au:Catalase Ratio = 130/32.5
    1. 500uL gold
    2. 407uL Hb
    3. 4093uL water
  7. Au:Catalase Ratio = 150/37.5
    1. 500uL gold
    2. 353uL Hb
    3. 4147uL water
  8. Au:Catalase Ratio = 170/42.5
    1. 500uL gold
    2. 312uL Hb
    3. 4188uL water
  9. Au:Catalase Ratio = 190/47.5
    1. 500uL gold
    2. 279uL Hb
    3. 4221uL water
  10. Au:Catalase Ratio = 210/52.5
    1. 500uL gold
    2. 252uL Hb
    3. 4248uL water

In order to get a more detailed view of the Catalase trends I made the following solutions:

  1. Au:Catalase Ratio = 7
    1. 500uL gold
    2. 2556uL catalase
    3. 1944uL water
  2. Au:Catalase Ratio = 17
    1. 500uL gold
    2. 1096uL catalase
    3. 3404uL water
  3. Au:Catalase Ratio = 26
    1. 500uL gold
    2. 697uL catalase
    3. 3803uL water
  4. Au:Catalase Ratio = 36
    1. 500uL gold
    2. 511uL catalase
    3. 3989uL water
  5. Au:Catalase Ratio = 46
    1. 500uL gold
    2. 404uL catalase
    3. 4096uL water
  6. Au:Catalase Ratio = 55
    1. 500uL gold
    2. 333uL catalase
    3. 4167uL water
  7. Au:Catalase Ratio = 65
    1. 500uL gold
    2. 284uL catalase
    3. 4216uL water
  8. Au:Catalase Ratio = 74
    1. 500uL gold
    2. 247uL catalase
    3. 4253uL water
  9. Au:Catalase Ratio = 84
    1. 500uL gold
    2. 219uL catalase
    3. 4281uL water
  10. Au:Catalase Ratio = 93
    1. 500uL gold
    2. 197uL catalase
    3. 4303uL water

I also took UVVis of the gold, Hb, and catalase stock solutions.

For each, I measured the spectrum for a 10X dilution of each stock solution.
