Objective

Copper catalyzed click reaction with azide flor

Description

Hydrogels

  • copper catalyzed click run with azide flor
  • dye in dmso, add a lithe and mix with film in water
  • add Cu(I) react for over 30 mins @ room temp
  • Cu(I) is unstable so add CUSO4 (Cu(II)) and ascorbic acid.

copper concentrations

  • biocon:0.375mmol cu(mecn)4PF5, 1.7 mol% cu with respect to total alkyne stirred at room temp for 2 days
  • nihms- 1888395: 0.05 mmol CuI (9.52mg).
  • PNAS- 2008-salic: From Edu staining section of methods: 0.5-1mM CuSO4, 1-60µM fluorescent azide (in 10-100mM stock in DMSO) + 50-100mM ascorbic acid, (added last, they made a 0.5 M stock solution in water)
  • PNAS-2006- Dietracric: 200MM triazle ligand, 50 mM alkyne tag: 100mg/ml Cu(I)Br
  • 1m hoding dye in DMS and leave overnight


Calculations for amount of FLOUR

  • 17.25µL Butyne-1-ol in the hydrogele
  • 17.24µL/70.09g/mol * 0.926g/mol= 2.28E-4 moles of butane-1-ol
  • rxn 1:1 however we want 1.5 excess
  • azide flour 488 mw: 563.45g/mol
    • 564:45g/mol *2.28E-4mol=0.128g*1.5= 0.193 grams
      • HOWEVER WE ONLY HAVE 5mg, not enough of the dye, so we will cut off a portion of the hydrogel so decided to take a piece off of it

Mass of whole hydrogel ~12.1219 piece~ 0.8810 g Percent of original film= 7.26% orginal amount of dye needed~0.193g new amount~ 0.014g
