Tasklist

1. Prepare 500 mL 50mM NaCl

  • (50 X 10-3M NaCl) * (58.44 g/mol) * (1 L/1000 mL) * 500 mL = 1.461 g NaCl for 500 mL

2. Prepare 1 L 50 mM Glycine Buffer at pH 3.5

  • 50 X 10-3M Glycine) * (75.07 g/mol) = 3.754 g in 1 L
  • Add 4 mL 1 M HCl to lower pH to 3.5

3. Prepare 25 mL 1 g/L Lysozyme

  • 0 .025 g Lysozyme in 25 mL

Dialysis

1. Dialyze 4 mL of Lysozyme stock solution (solution that was prepared in previous steps) in 3 in dialysis tube

  • 3500 g/mol MWCO tubing in approximately 150 mL of each "soak" solution
    • NaCl
    • Glycine
    • Pure Soak Water (HPLC Water)
  • 25k g/mol MWCO tubing in Glycine soak

2. Dialyze 4 mL of Lysozyme colloid solution (30:1 [Au]:[Lysozyme]) in 3500 g/mol MWCO tubing for glycine soak.

3. Beakers were labeled, covered with parafilm, and placed on a shaker at low speed overnight.

pH and Conductivity

  • Prepare 2x, 5x, 10x, 20x, 50x, 100x, 200x, and 500x dilutions of 50 mM Glycine and NaCl stock solutions.
  • The pH and conductivity measurements are summarized in the tables and graphs below.

NOTE The 0.1 mM solution was omitted because all of the data measured were outliers. The omitted data is shown in the table but is NOT included in the graphs. The outlier is indicated on the table by *.
