Purpose

  • To test the absorbance in solutions with various ratio of gold nanoparticles (AU) to BSA (Bovine Serum Albumin)
  • To discover trends in the increase of Au/BSA ratios

Procedure

  • Bovine Serum Albumin stock solution was prepared at 15.806uM.
  • Chloroauric Acid (HAuCl4) stock solution was prepared at 9.862mM.
  • The HAuCl4 stock wre measured by combining 0.0971g of HAuCl4 in 25mL of water. The BSA stock were calculated by combining 0.0105g of BSA in 1mL of water. The formula weight of HAuCl4 is 393.83g/mol and formula weight of bSA is 66430 g/mol. Please see Dhea's notebook for details of the calculation to determine the concentration of the two stock solutions.
  • BSA and chloroauric acid at the above concentrations was mixed in various amounts and was diluted with sterile water to the proper ratio. See table below for details.
Au/BSA ratio BSA[uL] HAuCl4[uL] H2O added[mL]
60 253.1 147.9 3.5990
80 253.1 197.2 3.5497
100 253.1 246.6 3.5004
120 253.1 295.9 3.4511
128 253.1 315.6 3.4313

160 253.1 394.5 3.3524
170 253.1 419.1 3.3278
  • 40mL of solution with various Au/BSA ratios were labeled and placed in incubator for four hours at 85°C, and left in room temperature until the following week.

Notes

  • All ratios of Au/BSA Solutions appeared yellow and transparent when mixed.
  • Test tubes with solutions enclosed were wrapped around with aluminum foil to prevent light-stimulated effects.
