*Ok so I checked the plates. I saw very small colonies on the both methods for pGreen, and small colonies on the TSS transformed colonies with my gene of interest. This is promising since if I didn't have ampiciliin there would already be colonies. So, I believe there is too much ampicillin, which shouldn't be a bad thing.
 
*Hopefully tomorrow the colonies will be bigger
