  • 57 fifth grade students


Banana scent demo: transformations

  • Banana: pTet-ATF1 (BBa_J45200) (Note: requires isoamyl alcohol to produce smell)
  • Amp resistant
  • Strain 1: BL21
  • Strain 2: DH5α-turbo
  1. BL21 + BBa_J45200
  2. BL21 neg.
  3. DH5α-turbo + BBa_J45200
  4. DH5α-turbo neg.


  • Transform with 0.5 uL DNA in 10 uL dH2O, or just water (neg)
  • Use 50 μL chem. competent cells for each
  • Plate on 100 μg/mL ampicillin
  • Grow o/n at 37°C to get colonies
