4:00-6:00. Finishing up the process, did a huge gel extraction (4g of gel!), over 10 tubes. PCR purified using the new PBI reagent. Nanodropped, got 18.1 for 335/sp and ~8 for 832/xp, with good 230/260 values; pretty confident that the DNA is there. Now waiting for the ligation. Put samples, marked with date, in our freezer; tossed old samples.
