Objective

  • to run UV-Vis spectra on the Au/BSA mixtures that Dr. Miller made on Friday,
  • to centrifuge the mixtures that Dr. Miller made,
  • to make 100mM Tris Buffer with pH 10 and pH 8, and
  • to remake Au/BSA mixtures with concentrations of 60-133 because no purple solutions were created in the other mixtures.

Description

  • UV-Vis
  1. UV-Vis was run on 1mL samples of water and each sample with the wavelength range of 700-200nm, looking for a peak around 550nm.
  2. The data points were collected and imported in Excel.
  3. The data was corrected for water and plotted to better visualize the absorbance.
  • Remaking of the Au/BSA Solutons
  1. The Au/BSA solutions were remade using the same protocol from last week, except that the metal spatula was wrapped in parafilm so that the metal from the spatula would not react with the gold. Concentrations of 60, 80, 100, 120, 128, 130, 132, and 133 were made.

Notes

  • Only the mixture with concentration of 80 Au/BSA was a homologous purple mixture. [made 8/30]
  • The stocks made today were made with the incorrect concentrations, so the reaction did not occur properly.

  • All the solutions from Dr Miller's batch from August 30. The solution was made with the parafilmed spatula. The solution spent 4 hours in a 85°C oven.

  • The 60 Au/BSA, 80 Au/BSA, 100 Au/BSA, and 120 Au/BSA solutions from Dr Miller's batch from August 30. The solution was made with the parafilmed spatula. THe solution spent 4 hours in a 85°C oven.

  • The 128 Au/BSA, 130 Au/BSA, 132 Au/BSA, and 133 Au/BSA solutions from Dr Miller's batch from August 30. The solution was made with the parafilmed spatula. THe solution spent 4 hours in a 85°C oven.

  • The 134 Au/BSA, 136 Au/BSA, 138 Au/BSA, 140 Au/BSA, 160 Au/BSA and 170 Au/BSA solutions from Dr Miller's batch from August 30. The solution was made with the parafilmed spatula. THe solution spent 4 hours in a 85°C oven.

  • All the solutions looked identical. The reactants did not react with each other, and the concentration ratios were off significantly.
