* Since the Oomycete gradient PCR didn't appear to say the problem was temperature (see below), I'm gonna try a nested approach to get amplifiable sequence for oomycete TRFLP in samples where there isn't much. So I used the DC6 primer in combo with OomR plus 5x25 ul of MgCl per 50 ul reaction. 35 cycles and 62 degree annealing. Will take 1 ul of this and apply it to OomF and OomR reaction, run under same conditions for a total of 70 cycles of amplification. Let see on gels tommorow how well this approach works.

  • Gonna start writing introduction to TVD paper (copy bits from previous TVD reports)
  • Get figures together for Australia report
  • Prepare Earth Alive figures
