Objective

To make HRP-AuNPs for enzymatic studies.

Procedure

Creating the Solutions of Gold and HRP

  1. For 30:1 Au:HRP solution, 5.10 mL HRP stock and 850.34 μL of gold stock were added to a 10 mL volumetric flask. The solution was diluted up to 10 mL with deionized water.
  2. For 60:1 Au:HRP solution, 2.56 mL HRP stock and 850.34 μL of gold stock were added to a 10 mL volumetric flask. The solution was diluted up to 10 mL with deionized water.
  3. For 90:1 Au:HRP solution, 1.71 mL HRP stock and 850.34 μL of gold stock were added to a 10 mL volumetric flask. The solution was diluted up to 10 mL with deionized water.
  4. For 120:1 Au:HRP solution, 1.28 mL HRP stock and 850.34 μL of gold stock were added to a 10 mL volumetric flask. The solution was diluted up to 10 mL with deionized water.

Synthesis of HRP-AuNPs

  1. The 10 mL solutions of gold and HRP were placed into four different test tubes.
  2. The test tubes were placed in the oven at 80°C for approximately 3 hours.
  • From 12:40pm to 4:00pm.

Notes

  • The stock solution of gold was 2.94 mM.
  • The stock solution of HRP was 16.27 μM.
