Golden Gate Assembly Confirmation

  • Growing three colonies of each plate in 6mL LB broth for 6 hrs.
  • Miniprep Plasmids and measure the concentration.
  • Concentrations are very low.
  • Cut the plasmids with SpeI and NotI, Not band were seen on the gel.
