I want to learn more about the tethering process and what each component does. This way if something goes wrong it will be easier to troubleshoot then if I didn't know. I'm sure this will be an ongoing project.

Components and Properties

  • anti-dig - needed for tethering
  • PBS
  • DNA - required for tethering
  • microspheres - required for tethering
  • BGB - thought to block surface and keep microspheres separated
  • popping buffer - for use with enzymes primarily, but used for generic tethering
    • EDTA - to sequester Mg++ ions
    • NaCl
    • Sodium Phosphate
    • Tween
