Well there are definitely bands, but who knows how much DNA I got. I will purify this stuff along with yesterdays product and then try the PCR again but with the program I did yesterday. Maybe there is something magical about that program and that extension time.
Gel 2
I am running the gel at 100V because Koch doesn't like my 160V escapades. The PCR products have been purified as well and condensed into 1 tube per type (so two tubes total).
This looks a little dimmer than normal. Also I don't know why the PCR band is longer than the 100bp band in the ladder. Lane designations:
- Ladder
- Cy3 stuff
- Cy5 stuff
Another PCR to come and gel analysis to come tomorrow.

Was there anything better about running at the lower voltage? If not, I'm not opposed to something that works. What does the manual for the Owl apparatus recommend?
It just occurred to me that the products look longer because of the labels. At this small length of DNA, I can imagine that the big floppy Cy3 or Cy5 labels make a difference.
