New page

==Purpose==
*Thermostable DNA ligases can be added to a PCR reaction to ligate extended oligos. This is used for some mutagenesis methods.

==Procurement==
*Examples for thermostable DNA ligases are Taq ligase [http://www.neb.com/nebecomm/products/productM0208.asp NEB] and the Ampligase [http://www.epibio.com/item.asp?ID=265 Epicentre].

==Use==
*If you want to ligate extended oligos in a PCR reaction it is important to have them phosphorilated at the 5'-end. Oligos will only be ligated when they are assembled on a compementary strand.
