You will determine whether the N15 protelomerase parts are functional or not. In your experiments, you'll cotransform protelomerase-expressing plasmid and a plasmid containing the tos site. Assuming you get stable replication, you'll miniprep and map the tos plasmid to determine whether it was linearized or not.
