Your goal is to determine the toxicity of parts and the expression of various devices within the set. Your toxicity assays are based on growth curves for bacteria containing Pbad-expressed parts. Your expression assays are based on ELISA experiments in which you express parts fused to peptide dual-tags using either BugBuster (to release all proteins) or deoxycholic acid (for periplasmic proteins). You then perform ELISA assays using antibodies, HRP detection, and a plate washer.
