colony pcr

  1. pick half of an isolated colony with a toothpick of pipet tip and inoculate 25 µl TE; store the plate parafilmed at 4ºC.
  2. boil for 5 min.
  3. spin down at 13k for 1 min.
  4. use 2 µl as template for subsequent pcr.
  5. run out on a gel with respective control.
  6. pick other half of the positive clones and inoculate liquid o/n culture for miniprep.
