- dishes
- prepare CaCl2 competent Rosetta pLys cells
- make LB-AMP/Chloro plates
- transform RNAp into freshly made competent cells
- make 4 M NaCl
- monitor cells in shaker and move to cold room when OD600 reaches 0.5
- start O/N culture of XL1-blue cells
- aliquot and freeze RNaseH1 protein
