Overview
Yeast nitrogen base without amino acids with agar with glucose for plates. This protocol does NOT leave any room for additives.
Reagents
- Yeast Nitrogen Base (without amino acids)
- Glucose (sterile 20% solution)
- Bacto Agar
- MilliQ H2O
Protocol
- Container 1:
- Begin with approx. 700ml MilliQ H2O in 2L bottle
- Add with constant stirring: 20g of Bacto Agar
- Bring up to 800ml with MilliQ H2O
- Autoclave
- Allow to cool (50°F)
- Container 2:
- Prepare 100ml solution of 20% Glucose
- Autoclave
- Allow to cool (50°F)
- Container 3:
- Begin with approx. 80ml MilliQ H2O
- Add with constant stirring 6.7g of Yeast Nitrogen base (without amino acids)
- Bring up to 100ml with MilliQ H2O
- Autoclave
- Allow to cool (50°F)
Combine contents of all 3 containers and mix well
