pH adjustment for LB medium
- Before autoclaving the LB medium:
- dip the pH-meter probe in the medium to monitor pH (wash probe with deionized water and calibrate pH-meter with 2 points when required);
- ajust pH with HCl, NaOH (or KOH). While handling strong acid/base, always wear suitable protections!
- wash the pH probe with deionized water and store it in KCl 3M.
- Autoclave the medium

- According to the M9 medium preparation protocol, mix:
- water,
- glycerol and
- M9 salts.
- Dip the pH-meter probe in the medium to monitor pH (wash probe with deionized water and calibrate pH-meter with 2 points when required);
- ajust pH with HCl, NaOH (or KOH). While handling strong acid/base, always wear suitable protections!
- wash the pH probe with deionized water and store it in KCl 3M.
- Autoclave the mix in a bottle.
- Equilibrate at room temperature and add the remaining ingredients (casamino acids, thiamine, CaCl2, MgSO4).
