It is believed that if kinesin comes in contact with glass, it denatures. Regardless if it denatures or not, all motility experiments will not work if the glass slides and slips are not passivated. To passivate a slide, the literature suggests using casein. In this buffer, we use [math]\displaystyle{ \kappa }[/math]-Casein as the surface passivator.

Recipe

  • PEM
  • 0.5 mg/mL [math]\displaystyle{ \kappa }[/math]-Casein

[math]\displaystyle{ \kappa }[/math]-Casein will go into solution relatively easily. You just need to let it stir on the stir plate for 30 minutes at a stir rate of 8. After the casein has completely gone into solution, filter it through a 0.2 µm syringe filter and aliquot it into your favorite tube/vial. It can be stored in the +4˚C refrigerator and in the e•IceBuckets.
