*4M potassium chloride (KCl)&lt;br /&gt;
*1M magnesium sulfate (MgSO&amp;lt;sub&amp;gt;4&amp;lt;/sub&amp;gt;)&lt;br /&gt;
*1% hexadecyltrimethylammonium bromide (CTAB)&lt;br /&gt;
*10% sodium deoxcholate&lt;br /&gt;

*10 mg ONPG&lt;br /&gt;
*27 &amp;amp;mu;L &amp;amp;beta;-mercaptoethanol&lt;br /&gt;

==Protocol==&lt;br /&gt;
#Grow cultures under whatever conditions you wish to test.&lt;br /&gt;
#During growth&lt;br /&gt;
##Make permeabilization solution.&lt;br /&gt;
##Pre-measure 80 &amp;amp;mu;L aliquots of permeabilization solution into 1.5 mL microfuge tubes and close them.&lt;br /&gt;
#Measure Abs&amp;lt;sub&amp;gt;600&amp;lt;/sub&amp;gt; of cultures and RECORD IT!&lt;br /&gt;
#Remove a 20 &amp;amp;mu;L aliquot of the culture and add it to the 80 &amp;amp;mu;L of permeabilization solution.&lt;br /&gt;
#*The sample is now stable for several hours.  This allows you to perform time-course experiments.&lt;br /&gt;
#*Also include a blank (solutions-only) sample for zero'ing the spec later.&lt;br /&gt;
#Make substrate solution.&lt;br /&gt;
#Warm samples and substrate solution to 30&amp;amp;deg;C&lt;br /&gt;
#Start timer counting up.&lt;br /&gt;
#Every 15 secs, add 600 &amp;amp;mu;L of substrate solution to a sample tube.&lt;br /&gt;
#Note the time of addition.&lt;br /&gt;
#After sufficient color has developed, add 700 &amp;amp;mu;L of 1M Na&amp;lt;sub&amp;gt;2&amp;lt;/sub&amp;gt;CO&amp;lt;sub&amp;gt;3&amp;lt;/sub&amp;gt;, mix well.  &lt;br /&gt;
#Note the stop time.&lt;br /&gt;
#Once all reactions are complete, transfer the tubes to a microfuge and spin for 10 minutes at full speed.&lt;br /&gt;
#Gently remove tubes from centrifuge.&lt;br /&gt;
#Measure the absorbance at 420nm and 550nm.  (Use UV-Vis protocol on [[Nanodrop]]).&lt;br /&gt;

* When is the reaction done?  I never found a good answer for this in the literature.  If I let a reaction go to completion, I measured an Abs&amp;lt;sub&amp;gt;420&amp;lt;/sub&amp;gt; of ~2-3.  Of course, this is out of the reliable range of the spectrophotometer, but it gives an indication of how far the reaction can go.  I got reproducible data when the yellow color was just detectable before adding the stop solution up to about the color of LB broth before stopping.  Remember, you need the substrate to saturate the enzyme during the course of the reaction, so don't let them go too far.  I routinely make three separate measurements for each culture and average them.&lt;br /&gt;
