The C-terminal domain of RNA pol II (RPII-CTD) is an unstructured tail fragment. It consists in multiple repeats of a heptapeptide repeat motif, which can be post-tranlationally modified by dedicated kinases ('writing'). These modifications are differentially recognized ('read') by a repertoire of PRMs across different homology families to regulate the activity of the multi-protein complex. Given knowledge of the recognition rules, designed chimeras can be combined to rewire the transcribing activity of RNA pol II, specifically, the elongation phase.
