To explore various methods of fluorescent detection of Calcium, we either added Fluo-3 AM or transfected inverse-pericam into mouse embryonic stem cells, then treated some cells with histamine and others with cyproheptadine hydrochloride (Calcium mobilizing agents). Here is shown MES cells + Fluo-3 AM + 0.1 uM cyproheptadine hydrochloride @ 10x magnification. We see increased levels of Calcium as a result of adding cyproheptadine (untreated MES cells + Fluo-3 AM show significantly less fluorescence).
