- plan to transform two plasmids, this afternoon into Top10, Bl21(DE3), KRX
- out of incubator tomorrow morning ready for overnight cultures on Monday
- grow Tuesday and overnight for KRX
- start purification on Wednesday
- keep things simple for Thursday
- gel filtration Friday

- have two colonies - might be the right thing
- recloning of sortase - redoing because got no colonies
- hope to be growing up next week
