  • Ligation (after digested all with X and P):
    • pSG001 as negative control
    • pSG002 as negative control
    • pSG005 (psG001+pRG006 (pLac from Antoine))
    • psG006 (pSG002+pSG004 (pTet from kit))
  • Transforming the ligation product and plating
  • Digesting pSG004 and pRG006 with X and P
