Competence test

  • Transformed PUK plasmid and YFP plasmid into mutant E.coli strains.

osmY and Kdp

  • digested PCR products, and death gene vector with EcoR1 and Pst1
  • ligated new BioBricks into vector
  • Transformed into competent TOP10 cells
  • Plate out onto Chloramphenicol plates
