Continued troubleshooting for LasR.

The dephos'd backbone + insert yields almost no colonies.
The dephos'd insert + gel purified backbone yields a lot of colonies.
This makes me think that something is up with the backbone. (The colonies on the plates are probably all the backbone re-ligating?)
I will try another method of obtaining the backbone; instead of gel purification, I can run a double digestion with E and X and heat deactivate.
