I checked the plates that were ligated yesterday, and there were colonies on the backbone control. There were no colonies on the negative control. So, we decided to colony PCR the 9 ligations performed yesterday. We picked 8 colonies per plate to PCR. We used the grid method to grow the colonies we picked for the PCR.
