I checked the plates that were ligated yesterday, and there were colonies on the backbone control. There were no colonies on the negative control.
AubI, BjaI, and SinI had a lot more colonies than the backbone only control plates. The other ligations varied in amount—most were similar to the amount that was on the bb only.

So, we decided to colony PCR the 9 ligations performed yesterday. We picked 8 colonies per plate to PCR.
We also picked colonies from the backbone control.

We used the grid method to grow the colonies we picked for the PCR.
After PCR, we plan to run the samples on a gel to see results.
