Golden gate colonies

  • 1-4 did not grow
    • Realized the Lux boxes were not correct. Ordered new ones
  • 5-6 grew >10 small colonies
    • Picked two from each plate into 3mL LB+amp
  • Positive control grew hundreds of medium sized colonies



Got some more K12s from Kylie to make competent cells. Picked more DH5αTs to make more competent cells
