EspA needle scFv

Monday 8-31: miniprep, restriction map, send for sequencing, transform plasmids into DH10B, plate
Tues 9-1: pick colonies into 10ml of media (do two for the two versions of the assays), grow for 2hrs to midlog, induce and grow for 4hrs til saturation (need to talk to Jenn), centrifuge and purify, grow up cultures for needle scFv w/control
Wednesday 9-2: carry out western blot (thurs?)
ask Chris about things to prepare for western blot. for instance, secondary antibody conjugated to fluorescent molecule

another way to carry out the assay: just like strep assay. requires the espA to be conjugated to fluorescent molecule
