Wet lab:

• Check plates from yesterday
• Start cultures (5ml) for wednesday of all biobricks

Dry lab:

• Finish colanic acid assay and SLIC cloning strategy

Results:

Wet lab:

• Checked plates from Monday - pBAD failed to grow so re-plated. Others appeared fine.
• Completed miniprep prep for Wednesday.

Dry lab:

• Continued (but did not finish colanic acid assay protocol).
• Read up on but did not design primers for SLIC.

Results:

• Did a RE and methylation assay, subject to approval
• Put up a cloning strategy to fit multiple cloning advantage of SLIC
• Looked through M1

• Helped Dineka and Kun with M3 assays.

Wet Lab

• Completed PCR of OtsB - failed to obtain bands on gel so this will be re-done.
