ProcedureSmart Phone Camera Settings

Calibration 1. Use the switch for the Blue LED to turn on the excitation light. 2. Check the settings on the smartphone and turn on the camera of your smart phone if they can be adjusted. a. Turn the flash. b. Set ISO to 800 or higher if possible. c. Set white balance to auto. d. Set exposure to highest setting. e. Set saturation to the highest setting. f. Set contrast to lowest setting. 3. Place your smart phone on the cradle at a right angle from the slide and adjust the height of the fluorimeter using the plastic trays so that your camera takes a picture of the drop sideways. This is very helpful because it makes filtering the blue light easier using ImageJ. 4. Adjust the distance between the smartphone on its cradle and the first two rows of the slide so that it is as close as you can get without having a blurry image. You should be at least 4 cm away from the drop. 5. Using the ruler provided in the lab, record the distance between the smart phone cradle and drop. Make sure not to move the camera, or fluorimeter very often. The light collected will change slightly if there is a significant difference from one image to the next in these distances.
