Splitting ARPE-19 cells

Before starting:

  • Warm up Media+Serum in 37°C water-bath
  • Place a beaker for disposing of waste in the hood
  • Place a T-25 Flask in the hood
  • Place 9 mL of warmed Media+Serum into a 15 mL conical


1. Thaw aliquot of cells in 37°C water-bath, making sure to avoid submerging the cap and o-ring; this should take about 2 minutes

2. Move cells into the 9 mL of media

3. Spin cells at 500 rpm for 3 minutes

4. Remove supernatant

5. Re-suspend the pellet in 5 mL of media

6. Place the cells into a T-25 flask
