* The controls for the transformation showed that the transformation worked properly, and that ampicillin resistance was being selected for.
* For the ligation controls, only two colonies grew which contained only the plasmid.
* The most efficient ligation was the 1:1 ratio, and plated in an 8μL quantity. This plate showed 50 colonies. Other colonies were observed on the 2:1 ratio.
* Eleven colonies from the 1:1 ligation and one from the 2:1 ligation were used to inoculate liquid cultures containing ampicillin. Incubated at 37°C , to be used tomorrow for plasmid extraction.

  • The controls for the transformation showed that the transformation worked properly, and that ampicillin resistance was being selected for.
  • For the ligation controls, only two colonies grew which contained only the plasmid.
  • The most efficient ligation was the 1:1 ratio, and plated in an 8μL quantity. This plate showed 50 colonies. Other colonies were observed on the 2:1 ratio.
  • Eleven colonies from the 1:1 ligation and one from the 2:1 ligation were used to inoculate liquid cultures containing ampicillin. Incubated at 37°C , to be used tomorrow for plasmid extraction.
