Confirmation of DNA content

  • On Tuesday, we ran gel electrophoresis on our now concentrated genomic DNA samples. Only sample 3 (from samples 1, 2, 3, and 5) produced a visible band. We did not feel confident that this was DNA, so today we ran digested and undigested samples from sample 3 to confirm its DNA content before we proceed. If sample 3 shows that it is liable to digestion, we will proceed with PCR. If not, we will return to our other samples and may also extract DNA from Serratia marcescens with a new extraction protocol.
