DNA Purification through Phenol/Chloroform Extraction and Ethanol PrecipitationToday we located two protocols for DNA purificaiton, through phenol/chloroform extraction and ethanol precipitation. We separated our sample of crude DNA into three test tubes in 100 microliter increments to attempt one phenol/chloroform extraction, one ethanol precipitation, and one phenol/chloroform extraction followed by an ethanol precipitation. One of the phenol/chloroform extractions went awry, inverting the organic and aqueous layers. This, coupled with the accidental discarding of the supposed organic layer, caused us to only do two experiments: one with the phenol/chloroform extraction only, the other with the phenol/chloroform extraction and ethanol precipitation. The extractions were finished, and 2.5 volumes of ethanol (200 microliters) in tube 1. The samples were frozen. |
